What is Proteinase K?
Proteinase K is a broad-spectrum serine protease, its crystallization and molecular structure indicate it belongs to subtilisin family and has no significant substrate specificity. Proteinase K‘s main cleavage site of is the peptide bond at the carboxyl end of the hydrophobic amino acid such as aliphatic or aromatic, which means that it can degrade a really wide range of proteins.
The proteinase K can be used in a variety of molecular biology, cell biology and otehr experiments.
Due to the outbreak and the global spread of the COVID-19 pandemic, the demand for nucleic acid testing has surged, and the supply of raw materials for testing reagents is also extremely tight, including proteinase K for nucleic acid extraction. At the same time, the particularity of proteinase K production makes major global manufacturers hardly to rapidly expand their production capacity in the short term. Therefore, proteinase K has been in short supply for a long time and even now, which has largely restricted the supply of nucleic acid diagnostic products for diagnostic companies worldwide.
In order to alleviate the supply pressure of proteinase K and provide sufficient raw material for the detection of COVID-19 virus, the strong R&D team of Healthgen Biotech used the world-leading protein purification platform (OryzPur) to quickly solve the problems such as long purification time, quality and high capacity in a short time. Healthgen Biotech recombinant Proteinase K (rProK) is a recombinant enzyme from Tritirachium album and is expressed in Pichia pastoris. It has the identical molecular structure and enzymatic characteristics as the native protein.
The advantages of rProK from Healthgen Biotech:
Fig 1. Comparison of activity detection between commercial rProK and Oryzogen rProK using casein as a substrate, they have comparable results.
Fig 2 & Fig 3. Using rProK in DNA & RNA extraction, the DNA and RNA were highly native, undamaged, which meant rProK was effective to remove DNase and RNase. (Fig 2 is DNA , Fig 3 is RNA.)
Wuhan Healthgen Biotechnology Corp.(Healthgen Biotech), a high-tech enterprise specializing in plant-derived pharmaceutical research and development, has world-leading technology platforms, rice endosperm protein expression platform (OryzHiExp) and protein purification platform (OryzPur), which have obtained a number of patents and constructed a ton-level capacity for plant-derived recombinant human serum albumin(OsrHSA), the first biological innovation drug of Healthgen Biotech. We make full use of the state-of-the-art technology platforms to provide customers with products of biopharmaceutical proteins and extraction and purification technology services of proteins that are in huge demand, but with the problems of no effecient platform expression or high purification system cost.
